RNA enzyme scavenger is a chemical modifier of RNA enzyme that reacts with imidazole ring of histidine, the active group of RNA enzyme, to inhibit enzyme activity; Protein, histidine modifier;
RNA enzyme scavenger properties:
It's fruity. Sensitive to moisture. Soluble in ethanol, ether, acetone and hydrocarbons, slowly soluble in water and hydrolysis, to produce ethanol and carbon dioxide. The relative density is 1012. The boiling point is 93 ~ 94℃ (2.40kPa). The refractive index is 1.395 ~ 1.398. The viscosity is 1.97mPa.s (20℃). Flash point 69℃. Low toxicity, median lethal dose i (rat, oral) 850mg/kg. It's stimulating.
RNA enzyme scavenger Uses:
Biochemical research, ribonuclease inhibitors. Protein, histidine modification reagent. Food fermentation inhibitor. Moderating esterification agent. Preservative for wine, soft drinks and fruit juices.
RNA enzyme scavenger notes:
1) RNA enzyme scavenger is irritating, stimulating the eyes, respiratory system and skin, the operation process as far as possible in the fume hood, its toxicity is not very strong, but the most toxic inhalation.
2) RNA enzyme scavenger is a potential carcinogen, mainly can produce ethyl ester derivatives.
3) RNA enzyme scavenger will produce carcinogens when mixed with ammonia solution, so be careful when used.
4) RNase scavenger can react with amines and sulfhydryl groups, so reagents containing Tris and DTT cannot be treated with RNase scavenger.
The difference between RNA enzyme scavenger water and RNA enzyme scavenger treated water:
RNA enzyme scavenger water refers to an aqueous solution containing RNA enzyme scavenger. Generally, 1ml of the original RNA enzyme scavenger solution is added to 1000ml of double steaming water, and then shaken overnight without high pressure.
Rnase scavenger treated water: that is, RNase free ddH2O, generally add 1ml of RNase scavenger stock solution into 1000ml double steamed water, then violently shake well, stand at room temperature for several hours, autoclave to remove degraded RNase scavenger (RNase scavenger is decomposed into carbon dioxide and ethanol).
Mechanism of RNase scavenger inhibiting nuclease:
RNA enzyme scavengers can bind to histidine in proteins and denature proteins. Rnase scavengers can also react with RNA or single-stranded DNA to destroy most of the adenine rings in single-stranded nucleic acids, but the concentration of destruction of single-stranded nucleic acids is 100 to 1000 times greater than the concentration of denaturation of proteins.
