he rTEV protease (Recombination type) is a genetically engineered recombinant protease that specifically recognizes the Glu-Asn-Leu-Tyr-Phe-Gln-Gly seven amino acid sequence. Compared to other proteases such as gut kinase U(EK), Thrombin, FactorXa, and SUMO, rTEV protease exhibits dual characteristics of high activity and specificity. Therefore, it is highly effective in removing fusion tags after expression of fusion proteins.The enzyme was purified by 6×His labeling (containing histamine label), with a purity of 99%. After the shear reaction, the enzyme can be removed by His labeling purification resin Ni-NTA Resin (Cat.No. P2010) to achieve the purpose of purification of the target protein.

Inhibitors
1) PMSF and AEBSF (1 mM), TLCK (1 mM), Bestatin (1 mg/ml), Pestatin A (1 mM), EDTA (1 mM) and E-64 (3 mg/ml), the above protease inhibitors had no effect on rTEV activity; 2) Zn ion (>5 mM) inhibited rTEV activity; 3) Reagents that react with cysteine are also potential inhibitors of rTEV.
