Product Introduction:

This product consists of 3 peptides and 2 low molecular weight proteins with a molecular weight range of 3.3 kd-20.1kd. It can be used to determine the molecular weight of peptides and small proteins on SDS-PAGE. This product is a freeze-dried powder of protein and polypeptide mixture, each protein content is about 15-22.5μg, equipped with a 1× loading buffer.

Gel preparation and dyeing precautions:

1. The separation glue is prepared first, and then the sandwich glue is prepared after polymerization, and the concentrated glue is prepared finally. The colloidal product ratio of the three kinds of glue is 4:1.5:1. After running at 30v for 1-2 hours, when the indicated front reaches the upper edge of the separation glue, the voltage is adjusted to 100v until the end of the electrophoresis, the whole electrophoresis process takes about 6-8 hours.

2. Due to the small number of amino acids contained in the polypeptide, if the polypeptide contains too many polar amino acids (basic or acidic), it will affect its band mobility on the SDS-PAGE chart, that is, its apparent molecular weight may be a certain distance from the amino acid theoretical molecular weight of the polypeptide.

3. Since the SDS-PAGE map shows that the molecular weight range of protein log-molecular weight and mobility is directly proportional to the linear relationship between them is 15,000-200,000, the molecular weight of proteins or peptides with molecular weight less than 10,000 can only be estimated according to the standard molecular weight to infer whether they fall into the predicted molecular weight range.

4. Due to the ultra-low molecular weight polypeptide (3000 and below), it is easy to leach from the gel, so the dyeing and decolorization time should not be too long, and the gel should not be soaked in water and preserved for too long after decolorization, otherwise the strip will disappear.

5. After electrophoresis, the glue can be fixed in the fixing solution for 20 minutes, and then dyed to obtain better protein bands; If time does not permit, do not fix direct dyeing. If dyeing with formula 7 is not effective or toxicity is considered, please choose our Coomas Bright blue protein glue Quick dyeing solution (Item No.:P1300), which is a fast, non-toxic and highly sensitive alternative to conventional dyeing solutions.
