Product Introduction
Compared with Bst2.0DNA polymerase and large fragments of BstDNA polymerase, Bst3.0DNA polymerase has better isothermal amplification activity and stronger reverse transcription activity. Whether using DNA or RNA as a template, this enzyme has 5 '-3' DNA polymerase activity and strong chain replacement activity, but the 5 '-3' and 3 '-5' exonuclease activity of this enzyme is missing.
In LAMP experiment using RNA as template, single enzyme system reaction can be realized. This enzyme has good reverse transcriptional activity between 60-65℃, which can effectively solve the reverse transcription of RNA templates with second-order complex structures, while Bst2.0DNA polymerase and large fragments of BstDNA polymerase have no such activity.
Precautions
(1) The used concentration of Mg2+ ranges from 4 to 10mM, and there is no Mg2+ in IsothermoBuffer. Generally, better LAMP results can be obtained under the condition of 6-8mMMg2+.
(2) It has been reported that adding TteUvrd helicase can improve the effect of LAMP.
(3) The specificity of amplification was tested using untemplated DNA as a control.
