Anti-fluorescence attenuation sealant is a kind of sealing reagent used to slow fluorescence attenuation. On the basis of conventional water-based sealing tablets, anti-fluorescence attenuation components are added to have a strong anti-fluorescence attenuation effect. It is used for sealing tissues and cell samples for fluorescence imaging. After sealing, the sample can be stored at 4℃ or -20℃ away from light for 2-3 weeks. The reagent is simple to operate, and it can be used to drop a drop of anti-fluorescence attenuation sealing solution with a pipette gun and cover the cover glass.
Anti-fluorescence attenuation tablets contain DAPI (4 ',6-diamidino-2-phenylindole), which is a fluorescent dye that binds strongly to DNA and is commonly observed with fluorescence microscopy. Because DAPI can penetrate intact cell membranes, it can be used for nuclear staining of both living and stationary cells, and a blue-fluorescent nucleus can be seen under the microscope. After the combination of DAPI and double-stranded DNA, the maximum excitation wavelength is 360nm and the maximum emission wavelength is 460nm.
