PDMPO is a fluorescent probe for lysosomal pH detection. Existing pH probes are not suitable for studying acidic organelles such as lysosomes, endosomes, phagosomes, sperm, and acrosomes because their fluorescence is significantly reduced at lower pH. While ratio imaging has received close attention over the past few decades, the growth potential of ratio imaging is limited by the lack of fluorescent probes suitable for acidic organelles. PDMPO features acid double excitation and double emission pH probes. It emits a strong yellow fluorescence at lower pH and a strong blue fluorescence at higher pH. This unique ph-dependent fluorescence makes PDMPO an ideal pH probe for acidic organelles with a pKa of 4.47. In addition, PDMPO has a large Stokes shift and excellent light stability, making it an excellent fluorescent acidic reagent for fluorescence imaging and flow cytometry applications. The unique fluorescent properties of PDMPO may provide researchers with a new tool to study cellular endocytosis, phagocytosis, and the acidic organelles of living cells. The UV laser for flow cytometry applications excites PDMPO well at 405 nm.
