Acid phosphatase ( ACP ) is widely present in various tissues, cells and body fluids. ACP catalyzes the hydrolysis of phosphate monoester to inorganic phosphate under acidic conditions, which is common in lysosomes of macrophages. ACP is often used in the auxiliary diagnosis of prostate cancer.
In an acidic environment, ACP catalyzes the hydrolysis of disodium phenyl phosphate to produce phenol, which reacts with 4-aminoantipyrine and potassium ferricyanide to produce red quinone derivatives with characteristic light absorption at 510 nm. ACP activity is calculated by measuring the increase rate of absorbance at 510 nm.
