Product Introduction:
Epoxy-Activated Beads 4FF are epoxy-activated agarose microspheres that can be used directly for the coupling of amino, sulfhydryl and hydroxyl proteins to samples. The pre-activated media can be prepared into special affinity media according to the needs, and the corresponding substances can be purified from the complex system in one step quickly and effectively.
Operating instructions:
1. Preparation of solution
Coupling solution: 0.1M Na2CO3, pH8.5-10.0
Sealing solution: 1M ethanolamine, pH8.0
Cleaning solution: 1.1M acetic acid-sodium acetate, 0.5M NaCl, pH4.0
Cleaning solution: 2.1M Tris-HCl, 0.5M NaCl, pH8.0
Preservation solution: 1XPBS containing 20% ethanol
2. Sample preparation
The sample was dissolved in a coupling solution at a concentration of about 5-10mg/ml.
3. Sample coupling
a. Take appropriate amount of Epoxy-Activated Beads 4FF, remove protective liquid, do not drain, clean three times with deionized water, and clean once with coupling liquid.
b. Dissolve dissolved samples and transfer them into cleaned Epoxy-Activated Beads 4FF. The volume ratio of filler: sample solution is about 1:1-2.
c, 25-40℃oscillation mixed reaction 24h. Note: Make sure the resin is suspended, otherwise it will greatly affect the coupling efficiency.
After the reaction, the coupling samples were collected to test the coupling efficiency. Clean the coupling solution once.
d, add equal volume of sealing liquid, 37 degrees oscillation mixed reaction 1h.
e. Remove the above reaction system, drain the solution, clean the resin with 3 times the column volume of deionized water, rinse the cleaning solution 1, deionized water, cleaning solution 2 and deionized water twice, and then store in the same volume of protective solution at 2℃-8℃.
