EdU (5-Ethynyl-2'-deoxyuridine) is a thymine nucleoside analogue, which can replace thymine to infiltrate into the replicating DNA molecule during the cell proliferation period, and the DNA replication activity of cells can be quickly detected through the specific reaction of EdU and Azide fluorescent dye, and the proliferation ability of cells can be detected. Compared with the BrdU detection method, the EdU detection method is faster, more sensitive, and more accurate. EdU is very similar to T, while EdU dye is only 1/500 of BrdU antibody, which is easy to diffuse in cells, without DNA denaturation (acid hydrolysis, pyrolysis, enzymatic hydrolysis, etc.), can avoid sample damage, and does not require antigen and antibody reactions, and can more accurately reflect DNA replication activity at the cellular and tissue level. This kit is suitable for value-added assays of cultured cells in vitro. Fluorescence detection should be used for adherent cells, which is suitable for the detection of fluorescence microscope, confocal microscope and high-content screening instrument. Suspension cells can be smeared after incubation with EdU, and the same staining procedure is applied to adherent cells from fixation after smearing.
