Bone tissue is divided into hard bone and cartilage. There are many methods for bone staining, such as toluidine blue method, Alizarin blue method, safranin O method, safranin O-fast green staining method, Goldner trichrome staining method, etc. Methylene Blue-Acid Fuchsin staining solution can clearly display osteoblasts, cell matrix, and osteoid.
After staining with methylene blue-acid fuchsin staining solution, the hydroxyapatite coating on the surface of the implant appears purple red, with clear display of osteoblasts, nuclei, and osteoids. The nuclei of osteoblasts are stained dark blue, the cell matrix is stained light blue, and the osteoids appear purple gray. The new bone tissue is dark red in color, with a clear boundary with the implant bone interface, while the mineralized bone tissue is red in color, with a blurred boundary with the newly formed bone.
