Mitochondrial Crimson Fluorescent Probe II is a cell-permeable mitochondrial crimson fluorescent probe that can specifically label biologically active mitochondria in cells and detect mitochondrial membrane potential. This product is a high-purity probe, which can be compatible with subsequent cell fixation and permeabilization, but after staining, the fluorescence intensity will decrease to a certain extent. This product is a deep red fluorescent dye that can be passively transported across the cell membrane by simple incubation with cells, and biologically active mitochondria are specifically labeled with the help of the mildly thiol-reactive chloromethyl functional group contained in this probe. The weakly sulfhydryl-reactive chloromethyl group contained in this probe can react with the sulfhydryl groups of mitochondrial proteins and be covalently linked, so when the following experiments are treated with aldehyde fixatives such as paraformaldehyde or formaldehyde, as well as the cell-permeable detergent Triton X-100, the fluorescent labeling of the mitochondria will not disappear, but may decrease to a certain extent (about 2-8 times). Because the excitation and emission spectra of this fluorescent probe do not typically overlap with common green and red fluorescent probes, it is ideal for use in multiplex fluorescence staining experiments.
This product can be used for mitochondrial fluorescence staining of live cells, and is not suitable for staining cells or tissues after fixation; However, after labeling mitochondria with this product, it can be fixed and permeabilized in the future, and stained with other probes or immunofluorescence staining, but after staining, the fluorescence intensity will be reduced to a certain extent. Moreover, the effects of fixation and permeabilization will be different for different cells, so it is recommended to perform certain tests on fixation and permeabilization reagents and conditions.
This product contains only a trace amount of powder, please centrifuge for a few seconds before use to allow the trace powder to settle to the bottom of the tube. Fluorochromes have quenching problems, please try to avoid light to slow down the fluorescence quenching.
