Hydroxyl radical is a kind of free radical which is highly toxic and harmful to organisms produced by human body in the process of metabolism. It can oxidize carbohydrates, amino acids, proteins, nucleic acids and other substances in tissues, suffer oxidative damage and destruction, leading to cell necrosis or mutation. Hydroxyl radical scavenging ability is one of the important indexes of antioxidant capacity of samples, which is widely used in the research of antioxidant health products and drugs.
H2O2/Fe2+ produced hydroxyl radicals by Fenton reaction. Hydroxyl radicals oxidized sulfonamides to diazo compounds under the action of electron acceptors, which are further coupled with naphthyl vinyl diamine. The red product had a characteristic absorption peak at 550 nm, and the absorption value was proportional to the content of hydroxyl radicals. The degree of color inhibition of the sample can reflect its ability to scavenge hydroxyl radicals. The kit is suitable for the determination of water-soluble samples, and BC1320/BC1325 is recommended for non-water-soluble samples.
