Product introduction:
Fluo-4 is a calcium fluorescent probe that replaces Cl with F in the Fluo-3 structure. By replacing Cl with F, which is more attractive to electrons, its maximum excitation wavelength will deviate about 10 nm to the short wavelength. This wavelength is closer to that of the argon laser, so Fluo-4 is twice as strong as Fluo-3 when excited with an argon laser.
Fluo-4, AM penetrates the cell membrane into the cell and is cleaved by intracellular esterase to form Fluo-4, which is trapped in the cell. Fluo-4 is almost non-fluorescent when it exists as a free ligand, but can produce strong fluorescence when it binds to intracellular calcium ions. The maximum excitation wavelength is 494nm, and the maximum emission wavelength is 516nm. The changes of intracellular calcium ion concentration can be detected by laser confocal microscopy or flow cytometry.
HBSS: no calcium, no magnesium, no phenol red
