IPTG was used as a placebo inducer and X-gal as a chromogenic substrate, and both were used for blue-white spot screening. IPTG can induce the synthesis of an amino-terminal fragment of β-galactosidase from the Lac operon DNA segment of the vector, which can achieve intragenic complementation (α-complementation) with the defective β-galactosidase encoded by the host cell. Bacteria achieving alpha complementation spread on medium containing X-gal chromogenic substrate formed blue colonies. The insertion of exogenous DNA into the polyclonal site of the plasmid can destroy the α complementation, and white colonies will be produced. IPTG is also commonly used as an inducer of recombinant protein expression in genetic engineering.
