Diaminobenzidine (DAB) is the chromogenic substrate of peroxidase (POD), and DAB is mainly used for immunoperoxidase method, which is suitable for immunoimprinting or immunohistochemical reaction of horseradish peroxidase HRP label. Peroxidase catalyzes the reaction of the substrate and produces brown precipitate at the reaction site. Immunoimprinting can show bands directly on the membrane. The color development time of the immunohistochemical specimen is generally 5-20 minutes at room temperature, and then the color development is observed under the microscope. After sufficient color development, the specimen should be dehydrated and sealed in time. The final product can be observed directly under a light microscope or treated with OsO4 to increase the electron density of the reaction product for electron microscope observation.
