Frozen section embedding agent is a water-soluble mixture of polyethylene glycol and polyvinyl alcohol, which has been widely used in immunohistochemistry laboratories to support tissue during frozen section to increase tissue continuity and reduce wrinkles and fragmentation. And because the mixture is water-soluble, it can be dissolved in water when bleaching the film, so the background dyeing will not be increased in the future dyeing.
Frozen section embedding agent Embedding process : The temperature of different tissue sections is different, and the temperature of the frozen microtome can be adjusted according to actual needs.
1. Remove the tissue blocks in sucrose solution and put them into the mixture of 20g /L sucrose and embedding agent at 1:1 volume ratio, and soak them for 2h at room temperature or soak them directly in the frozen section embedding agent for 2h at room temperature.
2. Transfer to the new frozen section embedding agent and soak at room temperature for 4h, replace with the new frozen section embedding agent and soak at room temperature for 6h.
3. The specimen was placed on a tray, sliced by a constant cold box microtome (generally -22℃) with a thickness of 10μm, affixed to the pre-treated slide, and stored in the refrigerator at -20℃.
