Product Introduction:
Enhanced DAB color development kit is a horseradish peroxidase (HRP) reaction kit that can be used for immunohistochemical color development, in situ hybridization color development or Western, Southern, Northern, EMSA membrane color development kit. DAB is a common substrate for horseradish peroxidase. Under the catalysis of horseradish peroxidase, DAB produces brown precipitate. The brown precipitate is insoluble in water and ethanol and can be used for normal dehydration sealing.
This product adopts special formula, high sensitivity, low background, good repeatability, stable storage, easy to use, suitable for blot, immunohistochemical and immunocytochemical, dot blot and biochip staining and color reaction.
Operation instructions (for reference only) :
1. For tissue sections or protein-imprinted membranes, after incubation with horseradish peroxidase (HRP) labeled antibodies or other forms of probes, wash 3-5 times with appropriate washing solution for 3-5 minutes each time. (See Note 1)
2. The solution A, B and C are evenly mixed according to the ratio of 1:1:18, that is, DAB working liquid, which is recommended to be used within 4h. (See Note 2)
3. Add an appropriate amount of DAB color developing working solution to the tissue section or film to ensure that the sample can be fully covered. (See Note 3)
4, room temperature incubation 1-30 minutes away from light, the color development time is too long can cause the background to increase, so it should be closely observed the color development process (generally 3-10 minutes is ideal), and in the background is shallow and reach the appropriate color development intensity with water rinsing to terminate the color reaction.
5, for tissue sections or cell samples, after the color reaction is terminated, it can be re-dyed with other dyes (commonly used hematoxylin staining solution). For the film, after the color reaction is terminated, it can be dried at room temperature and stored away from light.
