Determination of Significance:
Cytochrome P450 enzymes are a group of isozymes mainly found in the liver and play an important role in the metabolism of exogenous substances, especially drugs and poisons. Cytochrome P450 and cytochrome b5 are two heme proteins in the P450 enzyme system, and the change of their ratios is closely related to the metabolic activity of P450.
Measurement Principle:
After the oxidized cytochrome B5 is reduced by sodium dithionite, it has a maximum absorption peak at 424 nm, and the content of cytochrome B5 can be calculated by measuring the difference in absorbance values between 424 nm and 490 nm.
Self Provided:
Visible spectrophotometer/microplate reader, cryogenic centrifuge, ultracentrifuge, thermostatic incubator, mortar/homogenizer, adjustable pipette, microglass cuvette/96-well plate, ice, and distilled water.
