Product Introduction
The whole system of freeze-dried microspheres constant temperature amplification series of products makes the stability, ease of use and convenience of transportation of diagnostic reagents greatly improved. The whole system of constant temperature freeze-drying microspheres is prepared by a proprietary microsphere freeze-drying process, with uniform product type, non-crushing, long-term storage and avoiding cold chain transportation. For the development of diagnostic kits, the primer can be dried at the bottom of the EP reaction tube, and a freeze-dried microsphere can be added to become a freeze-dried product of the full amplification system, so the reagent performance is more stable, easy to store and transport.
The product is a freeze-dried microsphere product of the whole system, containing reaction buffer, Mg2+, dNTP, Bst 4.0 DNA/RNA polymerase, etc., which does not contain any dyes, and only needs to add primers and templates to perform nucleic acid isothermal amplification. Bst 4.0 DNA/RNA polymerase has RNA template-dependent polymerase activity (reverse transcription) and DNA-dependent polymerase activity, so it can be used for thermostatic amplification of both DNA and RNA samples. This product is an excellent reagent for LAMP and RT-LAMP amplification reactions.
This product is the base version of the Bst 4.0 reagent and does not contain any color developing dyes. The initial design goal was to perform LAMP visual detection with the OG orange Green color-changing tube (item No. A3828-21), but it can also be used to match other fluorescent dyes such as SYBR Green I, Eva Green, SYTO-9, etc. Note that this reagent does not support HNB color development, turbidity, pH color change.
Precautions
(1) As for the use of mineral oil, after preparing the LAMP reaction system, a drop of mineral oil can be added to cover the upper part of the reaction liquid to reduce aerosol pollution. Mineral oil does not affect the dissolution of OG dyes.
(2) When using other fluorescent dyes, the use concentration can be optimized by itself, generally speaking, referring to the supplier to provide a concentration of 0.25~0.8x, too high concentration of dyes may lead to amplification failure.
(3) This reagent does not support turbidity, pH discoloration, HNB discoloration reaction, do not try.
(4) Recommendations for LAMP finished reagents, Bst 4.0 Basic Bead freeze-dried ball and OG orange green color-changing tube are stable at room temperature and can be stored for a long time. The amplification primer was added to the bottom of the OG orange green color-changing tube, dried at 70℃, and then a freeze-dried ball was added to prepare the complete system detection reagent.
